1. Introduction

 

The Giardia(GIA) Antigen rapid test card (colloidal gold method) uses rapid immunochromatographic technology to detect Giardia antigen. After adding a sample from the feces to the sampling well, if there is Giardia antigen in the sample, it binds to the colloidal gold labeled anti-Giardia monoclonal antibody to form an antigen-antibody complex that moves along the chromatographic membrane, and then binds to the Giardia antibody on the testing line to display a wine red color. If there is no Giardia antigen in the sample, no color reaction will occur.

3. Test procedure

1) Use swab to collect freshly excreted fecal samples. Do not collect dry or viscous bulk fecal samples, because collecting dry feces sample will lead to collecting too much sample and appear false positive.

2) Insert the swab into the tube with sample buffer immediately, rotate the swab strongly on tube well for at least 10 times and mix the solution evenly, making the sample been dissolved in sample buffer fully. Press the swab on tube well above the liquid level, making the liquid been pressed out fully, discard swab.

3) If the fecal particles are large, they can be left to settle for 1-2 minutes.

4) Take out test card from sealed bag. Put it on flat surface, use dropper to draw upper-layer bright liquid, drop 4 drops (do not over adding) slowly, one by one, to the sample hole with “S” mark.

5) At this point, you will see a wine red liquid flowing through the observation window. If the liquid is not flowing, you can gently press the area between the "sampling hole" and the "observation window" to help the liquid flow.

6) Put the test card at room temperature for 5-10min, read the result, the result is invalid after 10 minutes.

 

4. Results judgment

 

1) Positive: Both on T Line and C line being seen purplish-red color reaction.

2) Negative: no color reaction on Test Line (T Line),only on Control Line(C Line) being seen purplish-red color reaction.

3) Invalid: In the observation window, there are no color lines in the control line area (C) and the testing line area (T); Or only color lines appear in the testing line area (T)., it means the operation is wrong or the test card is invalid, test again.

 

5. Note

1). Products that have expired or have damaged aluminum foil packaging bags cannot be used.

2). The test card must be used within 1 hour after removal.

3). Please use the accompanying sample buffer. Do not mix Sample buffer between different products or Sample buffer from different batches of the same product.

4). The collected sample should be tested immediately, avoid long time storage.

5). The collection of samples directly affects the accuracy of testing, so ensuring the correct, sufficient, and appropriate collection of samples. Excessive collection of fecal samples can affect the normal flow of fluids and lead to incorrect results (black bands appearing), while too few samples may result in false negative results. Therefore, please control the sampling amount according to the diagram. 

 

6). Avoiding bubbles when adding samples to the sample hole of the test card. When adding samples, please keep the front end of the straw about 1 cm away from the sample hole to ensure accurate adding samples; If the distance is too close, the drop volume may be reduced or the sample size may not be added due to the error in judging the number of drops, which will affect the test effect

7). Used test cards and samples should be considered as pollutants and must be properly disposed of in accordance with relevant local regulations.

 

6. Frequently Asked Questions

1) Limitation of the product

This kit is a qualitative screening reagent, can detect Giardia(GIA) antigen accurately, but if sample is not enough or concentration of antigen is lower than sensitivity, it may appear negative result. Doctors should make a diagnosis after considering all clinical symptoms. An accurate clinical diagnosis should not be based on a single test result.

 

2) The sensitivity and specificity of the product.

Compared with other similar rapid test cards, the sensitivity of this test card to detect Giardia antigen is>99%, and there is no cross reaction with symptoms similar to c0r0navirus, parvovirus, adenovirus, and canine distemper virus.
